DESCRIPTION

DryTech TEMPase Master Mix Green is a lyophilized and optimized version of the well-established liquidized TEMPase Hot Start DNA Polymerase Master Mix from Ampliqon. The lyophilized format allows shipment and storage without cooling. Shipment without cooling reduces both transportation cost and the impact on CO2.

DryTech TEMPase 5x Master Mix Green is easily reconstituted in the 5x DryTech Buffer Green.

DryTech TEMPase 5x Master Mix Green is composed of Ampliqon TEMPase Hot Start DNA Polymerase, the NH4buffer system, dNTPs, green loading dye, a density reagent and magnesium chloride. The included green loading dye and density reagent eliminates the need for addition of loading buffer and sample preparation before gel electrophoresis

Fewer handling steps lead to better reproducibility, which makes DryTech TEMPase 5x Master Mix Green suitable for standard PCR applications, screening and high throughput testing.

The included 5x DryTech Green Buffer contains a green loading dye plus a density reagent allowing direct loading. During electrophoresis, the green loading dye separates into two electrophoresis tracking dyes: orange G and xylene cyanol FF, migrating at ~100 bp and 1000 bp on a 1% agarose gels, respectively.

The reconstituted DryTech TEMPase 5x Master Mix Green can be stored at 4 °C for 3 months and at -20 °C for prolonged stability up to 6 months.

PERFORMANCE OF DRYTECH TEMPASE MASTER MIX

Performance of Ampliqon DryTech TEMPase Master Mix was evaluated on five different DNA targets varying in lengths. Each amplification has been conducted according to the recommendation in the datasheet. Lane 1: PAH (203 bp), Lane 2: CFTR (613 bp), Lane 3: BAIP (788 bp), Lane 4: PAH1963 (1963 bp) and Lane 5: CFTR3053 (3053 bp). M: Marker (Iqon PCR Ladder).

DIRECT GEL LOADING

DryTech TEMPase 5x Master Mix Green contains a green dye and a density reagent for direct loading of amplified PCR products onto the agarose gel. During electrophoresis, the green loading dye separates into two electrophoresis tracking dyes: orange G and xylene cyanol FF, migrating at approximately 100 bp and 1000 bp on a 1% agarose gels, respectively.
